As these two finally are escaping the gatekept realm, it’s time to explain what the two do and potential for height growth whilst on the two “supplements” to dumb this down, I will explain in simple terms, Forskolin (Aka a plant which is very crucial for activating adenylyl cyclase). Also another simple explanation explaining the PTHrP. - PTHrP binds to pth1r which inevitably stimulates AC to make cAMP… after the activation, a couple of processes will happen then resulting in beta arrestin to pull the receptor through clathrin coated pits. After a few more processes occur cAMP molecules bind to the tandem cyclic nucleotide, then eventually after another process be able to release phosphorylate. The SiKs get inhibited by PKA creating a binding site for 14-3-3 proteins which will move the SiKs into the cytoplasm to block there inhibitory effects on HDAC4/5, PP2A removes phosphate groups from HDAC4 to allow it to bind to the dna to block the MEF2 gene(mef2 increases maturation)
Once this process is complete CREB which is cAMP response element binding protein will also get phosphorylated by PKA will then bind directly to the SOX9 promoter and co recruit p300 for chromatin access for SOX9 and collagen 2l aggrecan. CREB also recruits p300 to the zip521 gene promoter to increase its mRNA which leads to more zip521 which further blocks maturation by binding and
inactivating runx2 which is the main maturation gene.
This Forskolin study was pretty basic and skipped some things but covered delayed growth plate closure, and chondrocyte proliferation.
Now I will talk about Icariin which is found in a herb which I will only include initials because of its strange name HGW. If you just look up what Icariin comes from you’ll find out.
icariin actually promotes a lot of beneficial things for the growth plate. it can increase signaling proteins linked to delayed growth plate closure like PTHrP and decrease proteins linked to growth plate closure like RunX2. Though it is good to note that this effects seems specific to the growth plate since it can increase Runx2 in osteoblast but decrease Runx2 in chondrocytes. Icariin promotes expression of osteoblast marker genes Runx2 and Id-1 in pre-osteoblasts and enhances BMSC renewal and osteogenic differentiation. it does this through activating BPM4 and also the BMP2/Smad4 signaling pathway, inducing ERk and JNK activation, increasingß-catenin mRNA and decreasing intracellular ROS levels.
Icariin increases expression of ALP and Coll while also stimulating mineralization of osteoblasts. Additionally it can also have anti-apoptotic effects by protecting osteoblasts from hypoxia induced cell death and glucocorticoid induced osteoporosis.
